STAT3，即信號(hào)轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄激活因子3，別名包括急性調(diào)節(jié)蛋白、信號(hào)轉(zhuǎn)導(dǎo)激活因子、亞油酸誘導(dǎo)蛋白等。STAT3是一種轉(zhuǎn)錄因子，廣泛存在于多種細(xì)胞類型中，由人體細(xì)胞自身的基因編碼。其作用機(jī)制主要是在細(xì)胞接收到外部信號(hào)，如細(xì)胞因子、生長(zhǎng)因子或其他刺激后，被激活的酪氨酸激酶磷酸化，形成二聚體，隨后轉(zhuǎn)移到細(xì)胞核內(nèi)調(diào)控特定基因的表達(dá)。STAT3的激活涉及JAK/STAT、IL-6/STAT3等信號(hào)通路，其中Janus激酶（JAK）首先被激活并磷酸化STAT3，導(dǎo)致其同源二聚化和核內(nèi)轉(zhuǎn)位。STAT3在生物學(xué)上的意義非常廣泛，它參與調(diào)節(jié)細(xì)胞的多種生理過(guò)程，包括細(xì)胞增殖、分化、遷移、凋亡以及免疫應(yīng)答等。STAT3的激活對(duì)于維持細(xì)胞穩(wěn)態(tài)和適應(yīng)外界變化至關(guān)重要。此外，STAT3在多種疾病的發(fā)生發(fā)展中也扮演著關(guān)鍵角色，尤其是在腫瘤發(fā)展中，STAT3的持續(xù)激活與腫瘤細(xì)胞的增殖、存活、血管生成和免疫逃逸等密切相關(guān)。因此，STAT3成為了癌癥治療中的一個(gè)重要靶點(diǎn)。在自身免疫性疾病、炎癥反應(yīng)和組織修復(fù)中，STAT3的調(diào)節(jié)作用同樣不容忽視，靶向STAT3的干預(yù)策略可能為這些疾病的治療提供新的途徑。
