SLC1A5全稱為溶質(zhì)載體家族1成員5,又稱ASCT2或ATB0,是一種中性氨基酸轉(zhuǎn)運蛋白,主要負責(zé)谷氨酰胺、丙氨酸等氨基酸的跨膜運輸。SLC1A5在腫瘤細胞中異常高表達,尤其在依賴谷氨酰胺代謝的癌細胞中,通過維持細胞內(nèi)氨基酸平衡和能量供應(yīng),促進腫瘤生長與增殖。SLC1A5活性與mTOR、Ras等調(diào)控細胞代謝與增殖的分子網(wǎng)絡(luò)密切相關(guān),在乳腺癌、肺癌、結(jié)直腸癌等多種惡性腫瘤中均觀察到表達水平升高。近年來,針對SLC1A5的抑制劑開發(fā)成為抗癌藥物研究熱點,例如小分子化合物V-9302已進入臨床前研究階段,通過阻斷谷氨酰胺攝取誘導(dǎo)癌細胞凋亡。SLC1A5還與神經(jīng)退行性疾病存在潛在關(guān)聯(lián),阿爾茨海默病患者腦脊液中ASCT2水平變化提示其可能參與神經(jīng)元代謝異常過程。當(dāng)前研究正探索將該蛋白作為生物標志物或聯(lián)合治療靶點的可能性。
