S100P

S100P,全稱為S100鈣結(jié)合蛋白P,是一種小分子鈣結(jié)合蛋白,因其最初在胎盤中發(fā)現(xiàn)而得名。該蛋白屬于S100家族,具有典型的兩個(gè)EF手型結(jié)構(gòu),廣泛參與細(xì)胞增殖、分化、細(xì)胞骨架構(gòu)成、免疫反應(yīng)以及細(xì)胞外基質(zhì)分泌活動(dòng)等過程。S100P蛋白在細(xì)胞核和細(xì)胞漿中均有表達(dá),并可分泌到細(xì)胞外。在細(xì)胞外,S100P能與RAGE的胞外配體結(jié)合,通過活化下游ERK/MAPK信號(hào)通路控制相關(guān)靶基因表達(dá),調(diào)節(jié)細(xì)胞增殖;在細(xì)胞內(nèi),S100P蛋白可以與Ezrin蛋白結(jié)合,參與細(xì)胞骨架的重構(gòu)而影響細(xì)胞粘附和運(yùn)動(dòng),還可以與泛素化通路CacyBP/SIP蛋白結(jié)合參與β-catenin的降解,實(shí)現(xiàn)對(duì)腫瘤生存和轉(zhuǎn)移的調(diào)控。S100P蛋白在多種腫瘤組織中均有明顯的高表達(dá),如膀胱移行細(xì)胞癌、非小細(xì)胞肺癌、胃癌、胰腺癌、乳腺癌、前列腺癌、結(jié)直腸癌、卵巢癌、宮頸癌等,并參與了細(xì)胞異常增生、細(xì)胞惡變、腫瘤細(xì)胞的運(yùn)動(dòng)性增強(qiáng)、浸潤力增強(qiáng)等腫瘤發(fā)生和發(fā)展的病理過程,與部分腫瘤的分期、預(yù)后及藥物敏感性相關(guān)。目前,S100P蛋白已被認(rèn)為是潛在的腫瘤標(biāo)志物,主要用于前列腺癌、膀胱癌等的輔助診斷。近年來,針對(duì)S100P蛋白的藥物研發(fā)也取得了一定的進(jìn)展,例如開發(fā)針對(duì)S100P蛋白的單克隆抗體用于腫瘤的診斷和治療。
