LRRC15

LRRC15,全稱富含亮氨酸重復(fù)序列的蛋白15,也被稱為LIB和hLib。該蛋白在多種實體瘤中高表達(dá),特別是在乳腺癌、頭頸部、肺癌、胰腺癌等腫瘤的基質(zhì)成纖維細(xì)胞上表達(dá),同時在間充質(zhì)來源的癌細(xì)胞亞群,如肉瘤、黑色素瘤、膠質(zhì)母細(xì)胞瘤中也有表達(dá)。LRRC15的表達(dá)受TGF-β調(diào)控,在活化的成纖維細(xì)胞和間充質(zhì)干細(xì)胞中誘導(dǎo)表達(dá)。作為治療LRRC15陽性基質(zhì)纖維增生癌癥或間充質(zhì)來源癌癥的靶點(diǎn),LRRC15已成為腫瘤治療領(lǐng)域的研究熱點(diǎn)。目前,多家藥企已開發(fā)出針對LRRC15的抗體偶聯(lián)藥物(ADC),如艾伯維的ABBV-085和第一三共的LRRC15 ADC專利藥物,均顯示出良好的臨床前療效。此外,LRRC15在雙特異性T細(xì)胞銜接器和放射性核素治療領(lǐng)域也展現(xiàn)出重要潛力,進(jìn)一步拓展了該靶點(diǎn)的治療應(yīng)用場景。
