殺傷細(xì)胞凝集素樣受體G1（KLRG1）又稱CLEC15A，是表達(dá)于自然殺傷細(xì)胞和T細(xì)胞表面的免疫抑制性受體。KLRG1通過識(shí)別E-鈣黏蛋白等配體傳遞抑制信號(hào)，參與調(diào)控免疫細(xì)胞活化閾值，在維持免疫穩(wěn)態(tài)中起重要作用。慢性病毒感染患者和腫瘤微環(huán)境中的T細(xì)胞常呈現(xiàn)KLRG1高表達(dá)，這種終末分化表型與細(xì)胞增殖能力下降密切相關(guān)，可能影響抗病毒或抗腫瘤免疫應(yīng)答。在自身免疫性疾病領(lǐng)域，KLRG1陽性調(diào)節(jié)性T細(xì)胞亞群顯示出更強(qiáng)的免疫抑制功能，其動(dòng)態(tài)變化與多發(fā)性硬化等疾病進(jìn)程存在關(guān)聯(lián)。近年針對(duì)免疫檢查點(diǎn)的藥物開發(fā)中，KLRG1作為潛在靶點(diǎn)受到關(guān)注，已有研究團(tuán)隊(duì)開發(fā)出阻斷性抗體，在黑色素瘤等動(dòng)物模型中觀察到聯(lián)合治療可增強(qiáng)PD-1抑制劑療效。但相較于成熟靶點(diǎn)，其轉(zhuǎn)化醫(yī)學(xué)研究仍處于早期階段，配體互作網(wǎng)絡(luò)與下游信號(hào)傳導(dǎo)的具體機(jī)制尚待深入解析。
