HAUS7

HAUS7,全稱為HAUS augmin like complex subunit 7,常見別名為26S proteasome-associated UCH interacting protein 1(UCHL5 interacting protein 1)。該蛋白在細(xì)胞分裂過程中發(fā)揮重要作用,是HAUS augmin-like復(fù)合物的一部分,參與有絲分裂紡錘體的組裝、中心體完整性的維持以及細(xì)胞分裂的完成。研究發(fā)現(xiàn),HAUS7在睪丸和卵巢中的表達(dá)水平最高,并在脾臟、胸腺、精巢等組織中也有檢測(cè)到。此外,HAUS7還與食道鱗狀細(xì)胞癌(ESCC)的發(fā)生和發(fā)展有關(guān),其與γ-微管蛋白的相互作用對(duì)ESCC細(xì)胞的生長(zhǎng)和存活至關(guān)重要。針對(duì)這一相互作用的治療抑制劑目前尚未出現(xiàn),但已有研究證實(shí),干擾這種相互作用可以有效抑制ESCC腫瘤的發(fā)生和發(fā)展。此外,HAUS7在神經(jīng)再生中也發(fā)揮重要作用,作為DOCK3的關(guān)鍵結(jié)合伴侶,通過調(diào)控微管組裝促進(jìn)軸突再生。在視神經(jīng)損傷后的軸突再生過程中,HAUS7的缺失會(huì)顯著抑制再生過程,這為青光眼等神經(jīng)退行性疾病的治療提供了新的靶點(diǎn)。目前,針對(duì)HAUS7的藥物研發(fā)主要集中在干擾其與γ-微管蛋白相互作用的治療抑制劑,以及增強(qiáng)HAUS7介導(dǎo)的微管穩(wěn)定性,以促進(jìn)神經(jīng)再生。
