Cd40l
CD40L，全稱為CD40配體（CD154），是一種II型跨膜蛋白，主要由活化的CD4+ T淋巴細(xì)胞表達(dá)。它通過(guò)與廣泛表達(dá)的CD40受體結(jié)合，在B細(xì)胞的活化、分化、成熟和凋亡中發(fā)揮關(guān)鍵作用。CD40L還參與免疫反應(yīng)的調(diào)節(jié)和維持，例如在生發(fā)中心反應(yīng)中促進(jìn)抗體類別轉(zhuǎn)換，以及通過(guò)激活樹突細(xì)胞分泌IL-12，推動(dòng)Th1型免疫應(yīng)答。此外，CD40L還與多種整合素結(jié)合，參與炎癥反應(yīng)和細(xì)胞因子分泌。CD40L的表達(dá)異常與高IgM綜合征等疾病相關(guān)。目前，針對(duì)CD40L的藥物研發(fā)主要集中在自身免疫性疾病和腫瘤領(lǐng)域，例如CD40L激動(dòng)劑和拮抗劑，以及新型分子治療策略。一些CD40L拮抗劑已進(jìn)入臨床試驗(yàn)，并在系統(tǒng)性紅斑狼瘡、多發(fā)性硬化癥等疾病中顯示出療效。
