ACSL5
ACSL5,全稱(chēng)為酰輔酶A合成酶長(zhǎng)鏈家族成員5,是一種參與脂肪酸代謝的關(guān)鍵酶。它催化長(zhǎng)鏈脂肪酸與輔酶A結(jié)合形成脂酰輔酶A,這一過(guò)程是脂肪酸代謝中的關(guān)鍵步驟,對(duì)能量供應(yīng)、細(xì)胞膜形成和信號(hào)傳導(dǎo)都至關(guān)重要。ACSL5蛋白在腸道、肝臟和脂肪組織中高表達(dá),其產(chǎn)物脂酰輔酶A參與多種生理過(guò)程。研究發(fā)現(xiàn),ACSL5基因突變可能導(dǎo)致脂代謝紊亂,與肥胖、胰島素抵抗和2型糖尿病等疾病相關(guān)。ACSL5過(guò)表達(dá)會(huì)促進(jìn)脂肪酸向甘油三酯轉(zhuǎn)化,加劇脂肪堆積,而敲低其表達(dá)則可能減少脂質(zhì)儲(chǔ)存并增強(qiáng)脂肪酸氧化。此外,ACSL5還與結(jié)直腸癌相關(guān),過(guò)表達(dá)時(shí)通過(guò)激活Wnt/β-catenin信號(hào)通路促進(jìn)腫瘤細(xì)胞增殖。目前,針對(duì)ACSL5靶點(diǎn)的藥物研發(fā)正在積極進(jìn)行中,例如諾和諾德公司引進(jìn)的肥胖候選藥物L(fēng)X9851,通過(guò)抑制肝臟中的ACSL5酶來(lái)調(diào)節(jié)脂肪代謝,并延緩胃排空,展現(xiàn)出良好的臨床應(yīng)用前景。

IHC image of CSB-PA891734HA01HU diluted at 1:600 and staining in paraffin-embedded human endometrial cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
